Is there a route that gets from the Yacht Club to Port Orleans or AK Lodge directly? I don't see how it could be done without changing busses.
The wedding has two pick-up points for people who don't want to drive.
What is the realistic drive time by car from Yacht to P.O.? I don't know which one.

There are no buses that run direct between any resorts. A formerly bus-driving marsupial with proper grammar comes to the rescue. Shorty is correct, there is no direct bus service, although you never know what you can get if you want to pay for it (and if you do, I suggest you ask for Bob from Carver, MA as your driver). I am sure there is a price at which you could charter a Disney Transport bus, but I know not what that is.
As for drive times, from the Yacht Club to anywhere is about a 10-15 minute drive at the most (if you are taking your time), and the drive from AKL to PO is roughly 10-12 minutes. If I was driving with no traffic problems, my drive time from AKL to PO would be about 10 minutes, and from YC to PO would be in the neighborhood of 7-8.

Thanks. I have some more details. Apparently it is one wedding guest shuttle stopping at both AK Lodge and Port Orleans. A middle stop at the Yacht Club shouldn't be a problem.

Your biggest time eater is Buena Vista Dr. AKL to PO 10 minutes tops. Use Osceola Parkway to Victory Way to BVD to Bonnet Creek Parkway.
From YC/BC to PO Use Epcot Resort Blvd to BVD to Bonnet Creek Parkway. 7-8 minutes if traffic and traffic lights cooperate.
I would be honored to drive you if requested and Disney Transport is arranged.
